Mazdutide

Also cited in the literature as IBI362 / LY3305677

An experimental peptide based on a natural gut hormone. Researchers studied it mainly in phase 2 and phase 3 trials in China.

Regulatory status: Not licensed as a medicine in the United Kingdom. It holds no MHRA marketing authorisation, has no approved indication, and is not approved for human or animal use.

What it is

Mazdutide is a laboratory-made version of oxyntomodulin, a hormone produced in the gut. It acts on GLP-1 and glucagon receptors.

Published studies also called it IBI362 or LY3305677.

How researchers think it works

Studies found that mazdutide activated both GLP-1 and glucagon receptors. Researchers linked these targets with blood sugar control and the way the body used energy.

Researchers designed the peptide around oxyntomodulin because that natural hormone also acts on both receptors.

What has research seen in people?

Trials, mostly run in China, reported reductions in body weight and improvements in blood sugar measures compared with placebo in adults with obesity or type 2 diabetes.

Stomach and bowel effects were again the most commonly reported problem.

Because nearly all participants were recruited in one country, researchers said it was unclear how far the findings carry across to other populations.

What studies have found

  • Phase 2 trials published in The Lancet Diabetes & Endocrinology and other journals studied adults with overweight, obesity or type 2 diabetes. Researchers reported changes in body weight and blood sugar compared with placebo.
  • Researchers also reported phase 3 results from trials in China. Published records described a regulatory review there.
  • Digestive problems were the most commonly reported side effects.

What we still do not know

  • Most participants were enrolled in China. Researchers said that more studies were needed to know whether the findings applied to other populations.
  • Little long-term evidence was available at the time of writing.
